What is Pierce's Disease?

Pierce's Disease is a bacterial infection, which is spread by bugs that feed on grapevines, particularly the "glassy winged sharpshooter." Grapevines that become infected with PD can quickly become sick and die.

Molecular characterization of the 9a5c isolate of Xylella fastidiosa by rep-PCR

  • Abstract: The purpose of this work was to characterize the 9a5c isolate of Xylella fastidiosa, that was selected for the genoma project started in 1997 by FAPESP and Fundecitrus in Sao Paulo, through DNA amplification by PCR, using primers corresponding to repetitive sequences present in the bacterial genome (REP, ERIC and BOX (rep-PCR)). The methodology was standardized to permit genetic diversity studies, as well as to evaluate the purity of X. fastidiosa isolates. The 9a5c isolate of X. fastidiosa, causal agent of Citrus Variegated Chlorosis (CVC) has been identified by DNA amplification with the especific primers CVC-1/272-2int; RST31/RST33 + CVC1/CVC2, which generated 500 bp, 733 bp and 270 bp fragments, respectively. The haplotypes obtained by rep-PCR showed bands with molecular weights ranging from aproximately 400 bp to 4.000 bp.
