Effects of micro-environmental conditions on organogenesis of Beni Izu grapes

  • Abstract: This study was carried out to determine optimum culture conditions for Beni Izu grapes in vitro. Three phototrophic conditions; auto, mixo, and heterotrophic were employed to investigate the possibility of micro-environmental control by CO2 enrichment and high PPF. MS medium containing 2.0 mgcntdotL-1 BA was the most favorable for shoot proliferation induced from shoot tip, and these shoots were subjected to different culture conditions. Plant growth was enhanced with increasing air exchange rates and could be grown even at 35degreeC. The maximal increase of fresh and dry weight was obtained when the plants were cultured under mixotrophic condition at higher PPF (140 mumolcntdotm-2cntdots-1), while remarkable increment of leaf area was observed when they were grown under autotrophic condition. It was observed that the leaf stomata of the plants grown under well-ventilated condition showed morphologically normal characteristics represented as a large number of small sized stomata and clear distinction between guard and subsidiary cells compared to those under poorly ventilated condition.
